APPLICATION OF HPLC FOR ANALYTICAL CONTROL OF LARGE SCALE SYNTHESIS OF (2R, 3S)-3-PHENYLISOSERINE HYDROCHLORIDE |

Abstract |
(2R, 3S)-3-phenylisoserine hydrochloride 1 is an important intermediate in the synthesis of Paclitaxel - an anticancer drug.
In NS-3 Department ICRI we elaborated technology of compound 1 in ½ technical scale (20 - 25 kg per year). The preparation of this compound was performed according to the six-step procedure. The aim of this study was elaboration of method and conditions for analysis of (2R, 3S)-3-phenylisoserine hydrochloride by high-performance liquid chromatography (HPLC). We tested two analytical columns: Zorbax Eclipse XDB-Ph and Zorbax Eclipse XDB - CN (250 mm x 4,6 mm, f-5m). The isocratic elution of separated compounds was carried out by mobile phase - methanol & sodium phosphate buffer 90:10 v/v, using constant flow rate 1,0 cm3/min. Chromatographic analysis was performed on a Merck LaChrom D-7000 Liquid Chromatograph (detector UV, l=210nm). The influence of solvent, pH and ionic strength of mobile phase on separation and peak symmetry was studied. |
