The glutathione antioxidant system in goat kids erythrocytes in the first week of life

Abstract

OBJECTIVE: The periparturient period and delivery may be effected on oxidative stress for pregnant female and newborn mammalian infants. In this period a newborns is exposed to the loss of balance between the production of reactive oxygen species and the capability of mechanisms protecting against the oxidation effects.

Glutathione (L--glutamyl-L-cysteinylglycine), which is present in all mammalian tissues, provides reducing capacity for several reactions and plays an important role in detoxification of hydrogen peroxide, other peroxides, free radicals and is a key factor in intracellular antioxidant defense mechanism. We postulated that it might modulate the adaptational response of the newborn to a higher oxygen concentration after birth. The aim of this study was to evaluate glutathione metabolism during the first week of life in goat kids (Capra hircus).

MATERIALS AND METHODS: The study was performed on 12 goat kids in the first week of life. The animals were kept in coops together with their mothers, were fed with colostrum and mother’s milk; they had free access to fodder (hay, straw) and water.

In whole blood we determined total GSH (tGSH), GSH and GSH disulfide (GSSG) concentrations, GSH/GSSG ratio (colorimetric method; Bioxytech GSH/GSSG-412 Assay) and the activites of the erythrocyte GSH reductase – GR (Bioxytech GR-340 Assay).

RESULTS: The contents of total GSH in whole blood in newborn goat kids ranged from 681,32 µM to 965,78 µM (782,52 ± 134,80 µM). We observed relatively high GSH/GSSG ratio (155,68 ± 0,64) and erythrocyte GSH concentration (772, 60 ± 133,11 µM); the concentration of GSH disulfide was low (4,96 ± 0,85 µM). Erythrocyte GSH reductase activity was low (3,65 ± 0,87 U/g Hb). In the presented study significant correlations were found between GR and tGSH, GSSG, GSH.

CONCLUSION: Relatively low concentration of GSSG in blood of goat kids and high GSH/GSSG ratio indicate on high metabolic capacity the glutatione antioxidant system in newborn goat kids in first week of life, though low erythrocyte GSH reductase activity.
