The effect of atherosclerosis obliterans on the ferroxidase activity of ceruloplasmin in presence of the raspberry seed extract.

Abstract

Ceruloplasmin (Cp) is a plasma protein that carries about 95% of the total copper. Multiple biochemical activities of Cp have been observed, including copper transport, oxidation of various amines, oxidation of Fe+2 to Fe+3, scavenging superoxide and the other reactive oxygen species. The ferroxidase activity of Cp prevents the generation of oxygen species through reduction of molecular oxygen to water.

Ceruloplasmin is an acute-phase plasma protein whose relative concentration and the ferroxidase activity increases two- threefold during inflamation, pregnancy, after traumatic injury, atherosclerosis.

Many endogenic (albumin, superoxide dismutase, Cp) and exogenic (vitamines) substances can play the protective role against the reactive oxygen species in the human body. The same role is performed by some components of dietary beverages and products such as red wine, tea, fruits rich in polyphenolic compounds. However, the interaction between polyphenolic compounds and Cp or the other antioxidants of blood plasma in human is still unknown.

The aim of the study was to investigate the ferroxidase activity of Cp isolated and purified from the healthy subject serum (control group) and from patients with atherosclerosis obliterans (AO). In second step the effect of raspberry seed extract (RSE) on the ferroxidase activity of both of ceruloplasmin was analyzed.

The ferroxidase activity of Cp and the effect of raspberry seed extract was determined by spectrophotometry using Fe(II) – histidine complex and ferrozine as a chromogenic reagent.

The effect of AO on the ferroxidase activity of Cp, measured as the decrease of Fe(II) concentration in samples and expressed as ΔFe(II), was observed. For 0.139 [mg/ml] of Cp, ΔFe(II) was higher in AO with comparison to the control group (1.42 and 0.46 [μmol/l], respectively). The RSE demonstrated the ferroxidase activity within the studied range of 0.625 to 12.5 [μg d.m/ml]. The effect of RSE on the ferroxidase activity of ceruloplasmin both from AO and control group was observed in dose–dependent way. RSE affected the ferroxidase activity of Cp from AO more efficiently than that from the control group. However, the addition of the higher concentrations of raspberry seed extract (12.5 [μg d.m/ml]) to samples with constant amount of Cp promoted Fe(II) oxidation with a lower efficiency in both groups. It may be suggested that the equilibrium of reaction is reached and the addition of the higher amounts of both of antioxidants cause any change in Fe(II) elimination because of competition of antioxidants for the substrate.
