Human β‑casomorphin transport across Caco-2 monolayer |

Abstract |
The bioavailability of nutrients is an important factor of the health-nutrition interactions assessment. Direct in vivo experimentation on the intestine or its epithelium is difficult, thus in vitro cell culture models were developed. One of the commonly used cell line is Caco-2 – derived from the human colorectal adenocarcinoma, it has epithelial morphology and upon reaching confluence, expresses characteristics of enterocytic differentiation. It is used with in vitro assays to assess intestinal transport of various compounds, e.g. nutrients or drug candidates across intestinal epithelial cell barrier. The aim of our research was to determine determination of absorption ratio of human β‑casomorphin-5 and -7 transport in Caco-2 monolayer. Caco-2 was obtained from American Type Culture Collection and cultured in DMEM with 10% of FBS, 1% of NEAA and 50mg/dm3 gentamycin, incubated at 37°C in humidified atmosphere of 5% CO2 in air. Peptide transport was analyzed in apical and basal direction for 60 minutes of the experiment. Diprotin A was used as a dipeptidylpeptidase IV (DPPIV) inhibitor. Both investigated peptides (β‑casomorphin-5 and –7) were demonstrated to be capable of penetrating Caco-2 monolayer. Under conditions of reduced DPPIV activity, the transport of both casomorphins was increased. Kinetic analysis of the transport of β‑casomorphins across Caco-2 monolayer showed that the investigated peptides were transported mainly by passive mechanisms.
