Mitigating effects of triphala on fluoride non-endocrine toxicity.

Abstract

Healthy adult female rats (Rattus norvegicus) were administered sodium fluoride (NaF) at the dose of 10 mg/kg body weight and triphala at 30 mg/kg body weight orally. Both doses were given individually and also in combination for 30 days to investigate the non-endocrine organ fluoride toxicity in muscle and blood with reference to energy metabolism and its reversal by triphala. Fluoride caused an alteration in body and organ weights and decreased total protein levels in both gastrocnemius muscle and serum of NaF treated rats. The treatment also resulted in a reduction in phosphorylase and succinate dehydrogenase (SDH) activities followed by an increase in glycogen levels probably affecting energy and carbohydrate metabolism. This is further supported by increasing fluoride levels in the muscle tissue. The SGPT and SGOT levels were elevated by fluoride treatment indicating impairment of liver functions. Feeding also brought about a significant decline in hemoglobin level, mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin (MCHC) and R .B.C which suggested occurrence of anemia and probable effect on haemopoetic system. Significant increase in erythrocyte sedimentation rate (ESR) indicates toxic effects in the body. Increase in white blood cells count significantly suggests adverse effects of NaF treatment on the immune system of the rats. Co-administration of triphala to fluoride treated rats resulted in a partial effects in muscle and blood including body weight due to its probable protective role. Thus, triphala mitigated NaF induced non-endocrine toxicity marginally in a rat model.
