Reduced expression of the ZAC mRNA in bladder cancer |

Abstract |
The ZAC gene encodes a novel zinc finger protein that regulates apoptosis and cell cycle arrest and maps to chromosome 6q24-q25, a region known to harbor putative tumor suppressor genes frequently involved in solid cancer tumors. ZAC shares a number of similar functions to p53 tumor suppressor, and Zac protein is thought to act synergistically with the p53 in causing apoptosis, possibly by co-activation of p53 target genes. The purpose of this study was to investigate the expression of ZAC gene in bladder cancer samples using real-time RT-PCR technique. ZAC expression was measured in 26 frozen neoplastic and adjacent non-neoplastic tissues, paired of each patient. The analysis revealed that ZAC mRNA expression was reduced in 24 of 26 (92%) cases of bladder cancer. Greater decrease of the ZAC mRNA expression was observed in superficial (Ta/T1) tumors comparing with muscle invasive (T2-T4) bladder tumors. The altered expression of ZAC support the gene's potential role in the pathogenesis of bladder cancer. These results indicate that the decreased expression of ZAC gene is a very common and early event in bladder cancer. |
