Maize dwarf mosaic virus is a casusal agent of maize disease worldwide. It is a member of the Potyviridae family and Potyvirus genus, transmitted in non-persist manner by several aphid species. It infects many wild and cultivated grasses. At first time MDMV was detected at the beginning of the 1960s by Williams and Alexander in USA. In Europe the presence of virus was confirmed in Spain, France, Italy, Germany, Czech Republic. In Poland, for the first time MDMV was detected by ELISA test in 2005.

MDMV genom consists of positive-sense ssRNA encodes polyprotein which results in ten functionally potyviral proteins. The most useful object for serological and molecular virus detection is coat protein (CP).

The aim of our work was to analyse sequence of coat protein gene and comparison with other known MDMV sequences.

We studied two Polish MDMV maize isolates. Plant samples were collected from Southern and South-western part of Poland. Total RNAs extracted from fresh infected maize plant leaves were used as template. RT-PCR reactions were performed with literature primer pair specific for CP and NIb proteins. Amplified product consisted of a part of the NIb and a part of the CP. It was separated electrophoretically and isolated from the gel. Subsequently the DNA fragment was cloned and sequenced. Obtained sequence was analysed by BlastN program and manually edited in GeneDoc. The phylogenetic analysis was performed using PAUP 4.0 software.

Sequence comparison of partial coat protein gene showed homology of Polish MDMV isolates. Two our isolates indicated high identity of CP nucleid acid sequences with different Spanish, Argentine, Israeli, Hungarian and Bulgarian MDMV isolates.
