Antioxidative effect of the short-term vitamin E and C combined supplementation in men with different risk of atherosclerosis.

Abstract

Introduction: The large, clinical, prospective trials did not prove the efficacy of vitamin E supplementation in protection against oxidative stress and atherosclerosis. It has been suggested that the mode of vitamin supplementation (on empty stomach or with meals), as well as the patient inclusion criteria, may be related to the antioxidant vitamin bioavailability and effectiveness. The aim of the study was to evaluate the influence of the two-week, vitamin E (2 x 100 IU daily) plus C (2 x 200mg daily) supplementation on redox plasma status in: low-risk group of healthy men (n=23), moderate-risk group of men with ≥2 classical risk factors (n=39) and PVD group of patients (n=50); and to check, if the antioxidative effect of vitamin E plus C supplementation depends on: mode of vitamin ingestion, initial plasma redox status, and plasma level of new biochemical atherosclerosis risk markers -hsCRP and ischemia modified albumin (IMA). Methods: The modality of food intake was checked in the cross-over study. Plasma parameters: LDL oxidative susceptibility, TBARS, lipid hydroperoxides, vitamin E, vitamin C, thiol/albumin ratio, FRAP, hsCRP and IMA were measured. Results: The vitamin E plus C supplementation with meals augmented their bioavailability and the antioxidative effect in plasma. The effect was more evident in patients with existing risk factors or PVD than in healthy subjects. Conclusion: Food intake increases bioavailability of the antioxidant vitamins. More beneficial effect of vitamin E plus C was obtained in subjects with already existing decreased plasma antioxidants level and increased hsCRP and IMA concentration in plasma.
