The influence of non-oxidized and oxidized rapeseed oil with alpha-lipoic acid addition on lipids profile and paraoxonase-1 activity in rats

Abstract

Consumption of high-oxidized oil may by disturbing lipids profile and antioxidant enzymes activity influences on development of hypercholesterolemia. While supplementation the diet with antixodants may inhibit those changes. Alpha-lipoic acid is an antioxidant, and is widely used in prevention of various diseases such as atherosclerosis, diabetes or cancer. From second side, low activity of paraoxonase-1 (PON-1) is linked with clinically evident atherosclerosis. The aim of the study was to find out if the consumption of oxidized oil aggravates the undesired changes in the concentration of lipids and PON-1 activity in blood serum and whether the addition of ALA to diet results in anti-atherosclerotic action. The experiment were carried out on 30 rats divided into 5 groups: control group on standard diet and 4 study groups which received suitably: non-oxidized rapeseed oil, oxidized rapeseed oil, alpha-lipoic acid (ALA) and oxidized rapeseed oil with ALA addition. The rapeseed oil was oxidized for 8 hours at 1800C. Oxidation process caused an increased content of palmitic and oleic acid and a decreased content of linolic and linolenic acid. The experiment lasted 12 weeks. The body weight of the rats was checked once a week and after experiment blood samples were taken. Changes in the lipids concentration and PON-1 activity showed not only that oxidized rapeseed oil, administrated to rats caused the decreasing of HDL-CH concentration and PON-1 activity but as well that ALA addition didn’t influence beneficial on those changes.
